چکیده

Protamines are expressed in the spermatid nucleus and allow denser packaging of DNA compared with histones. Disruption of the coding sequence of one allele of either protamine 1 (Prm1) or Prm2 results in failure to produce offspring, although sperm with disrupted Prm1 or Prm2 alleles are produced. Here, we produced Prm1-deficient female chimeric mice carrying Prm1-deficient oocytes. These mice successfully produced Prm1(+/-) male mice. Healthy Prm1(+/-) offspring were then produced by transferring blastocysts obtained via in vitro fertilization using zona-free oocytes and sperm from Prm1(+/-) mice. This result suggests that sperm lacking Prm1 can generate offspring despite being abnormally shaped and having destabilised DNA, decondensed chromatin and a reduction in mitochondrial membrane potential. Nevertheless, these mice showed little derangement of expression profiles.

Re-visiting the Protamine-2 locus: deletion, but not haploinsufficiency, renders male mice infertile

Protamines are arginine-rich DNA-binding proteins that replace histones in elongating spermatids. This leads to hypercondensation of chromatin and ensures physiological sperm morphology, thereby protecting DNA integrity. In mice and humans, two protamines, protamine-1 (Prm1) and protamine-2 (Prm2) are expressed in a species-specific ratio. In humans, alterations of this PRM1/PRM2 ratio is assoc...

The effects of exposure to fluoxetine during lactation on testicular tissue and sperm parameters in mice offspring

Fluoxetine is a selective serotonin reuptake inhibitor is commonly prescribed to treat maternal depression in pregnancy and lactation. This study aimed to investigate the effects of maternal exposure to fluoxetine via lactation on testicular tissue, sperm parameters including count, motility, viability, and normal morphology and testicular oxidative stress status in male mice offspring. Ten mic...

The Effects of Protamine Deficiency in Sperm Humanon Premature Chronosomal Condensation

Purpose: After aneuploidy, failure of oocyte activation, resulting in premature chromosomal condensation (PCC), is considered as the second cause of failed fertilization post ICSI. Oocyte quality has been shown to affect PCC formation, and since this study sought to evaluate the effect of sperm chromatin anomalies on the PCC, mice metaphase II oocytes were used. Materials and Methods: Metaphas...

Sirt1 Deficiency Attenuates Spermatogenesis and Germ Cell Function

In mammals, Sirt1, a member of the sirtuin family of proteins, functions as a nicotinamide adenine dinucleotide-dependent protein deactylase, and has important physiological roles, including the regulation of glucose metabolism, cell survival, and mitochondrial respiration. The initial investigations of Sirt1 deficient mice have revealed a phenotype that includes a reduced lifespan, small size,...

O-10: Formation and Molecular Composition of The Sperm Head to Tail Coupling Apparatus

Background According to a worldwide survey in 2010 infertility affects 48.5 million of couples. In roughly half of the cases infertility is provoked by the male mate. Thus, a significant percentage of young men are infertile but the underlying causes are mostly unknown. Male fertility and reproduction success critically depends on proper formation of the mature sperm. Transmission of the geneti...
